Foundayo® (orforglipron) is a prescription weight loss medicine for eligible adults with obesity or overweight. It works by influencing appetite regulation, which may help you eat less and reduce your body weight. It is used alongside a reduced-calorie diet and increased physical activity.

How Does Foundayo® Help with Weight Loss?
Foundayo® contains the active ingredient orforglipron. It influences the regulation of appetite, which can help reduce food intake and support weight loss.
Foundayo® should be used alongside a reduced-calorie diet and increased physical activity as part of an overall weight management programme.
Who May Be Suitable for Foundayo®?
Foundayo® may be considered for adults who initially have:

A BMI of 30 kg/m² or above (obesity), or
-
A BMI of at least 27 kg/m² but below 30 kg/m² (overweight) together with at least one weight-related health condition.
Weight-related conditions may include prediabetes, type 2 diabetes, high blood pressure, abnormal blood fats, obstructive sleep apnoea, impaired heart function or a history of certain cardiovascular conditions.
Suitability is not determined by BMI alone. A clinical assessment is required before treatment is prescribed.
Is Foundayo® a Tablet or an Injection?
Foundayo® is a once-daily tablet containing orforglipron.
Unlike injectable GLP-1 weight loss medicines, Foundayo® is taken by mouth and can be taken at any time of the day, with or without food.
The tablet should be swallowed whole and should not be broken, crushed or chewed.
How Is Foundayo® Taken?
Treatment starts at a low dose and is gradually increased.
The starting dose is 0.8 mg once daily for 30 days. The dose is then increased to 2.5 mg once daily.
After at least another 30 days, the dose may be increased to 5.5 mg once daily.
If required, further increases may be made through 9 mg, 14.5 mg and 17.2 mg once daily, with at least 30 days at each dose level before increasing.
The maximum recommended dose is 17.2 mg once daily.
Your dose should only be changed following advice from your prescribing clinician.
What Are the Common Side Effects of Foundayo®?
The most commonly reported side effects include:

Nausea

Constipation

Diarrhoea

Vomiting

Indigestion

Abdominal pain
Nausea, diarrhoea and vomiting are particularly common when starting treatment and tend to reduce over time for many patients.
Other side effects can occur, which will be discussed as part of your clinical assessment and treatment monitoring.
Are There Serious Side Effects?
Although uncommon, serious side effects can occur.
Acute pancreatitis has been reported and may cause severe and persistent abdominal pain which can spread to the back.
Other important problems reported with this type of medicine include serious allergic reactions, bowel obstruction, severe constipation, kidney problems and certain visual symptoms.
You should seek appropriate medical attention if you develop symptoms suggestive of a serious adverse reaction.
Who Should Take Extra Care with Foundayo®?
Before treatment, your clinician needs to know if you have or have previously experienced conditions including:

Pancreatitis

Severe problems with digestion or severe gastroparesis

Gallbladder problems
-
Diabetic retinopathy or other relevant eye problems

Severe heart failure
-
Kidney problems
-
Planned surgery requiring anaesthesia
Your complete medical history will therefore be reviewed before prescribing.
Does Foundayo® Interact with Other Medicines?
Foundayo® can interact with other medicines.
Examples include certain antibiotics, antifungal medicines, antiviral medicines, epilepsy treatments and medicines used for cholesterol.
Particular consideration is also required when Foundayo® is used alongside insulin or sulfonylureas because the risk of low blood glucose may increase.
You should therefore provide your clinician with an accurate and complete list of prescription medicines, over-the-counter medicines and herbal products before treatment.
Foundayo® and Pregnancy
Foundayo® should not be used during pregnancy.
If you are planning a pregnancy, treatment should be discussed with your clinician. The patient information leaflet advises stopping Foundayo® at least 3 weeks before attempting to become pregnant.
Orforglipron may also reduce the effectiveness of oral hormonal contraception. Additional or alternative contraception may therefore be required for 30 days after starting Foundayo® and for 30 days following each increase in dose.
Foundayo® should not be used during breastfeeding.
